Substrate selection and oxygen uptake by the lactating mammary gland.

چکیده

Toutes les proteines et tout le lactose, et environ 50% des acides gras des lipides du lait sont synthdtisks par les cellules CpithCliales de la gland mammaire. Au pic de la lactation, il y a une demande CnergCtique ClevCe auprbs de ces cellules pour fournir les substrats circulants (glucose, acides gras, corps cCtoniques, triacylglycCrols et acides aminks), pour les rCactions de biosynthbse, et pour la sCcrCtion de lait. Le coDt d’energie individuelle le plus ClevC est imputC B la synthbse des 1actoprotCines. Cette Cnergie provient en grande partie de l’oxydation du glucose (chez les non-ruminants), et du glucose plus acCtate (chez les ruminants). Comme on peut s’y attendre, les taux mesur6s de l’extraction d’O2 par la glande en lactation chez le rat in vivo sont plus ClevCs (d’environ 50%) que ceux requis pour les rkactions purement de synthbse, et l’absorption d’O2 reste importante lorsque la production de lait est faible (comme chez le rat B l’etat de jeDne). Normalement, le principal prkcurseur de la lipogenbse est le glucose chez le nonruminant, YacCtate et le 3-hydroxybutyrate chez le ruminant. Le glucose est aussi le prCcurseur du lactose et du glydride-glydrol. Cependant, la consommation de substrats par le tissu mammaire est trbs sensible aux modifications de l’alimentation (un rCgime riche en glucides opposC B un rCgime riche en graisses, le jeDne), et aux hormones plasmiques (dCficience en prolactine ou en insuline). Nous discuterons ici les facteurs susceptibles de contrdler la consommation et la selection des substrats par la glande mammaire (y compris le fluf; sanguin du tissu).
